Nanofibrous Formulation of Cyclodextrin Stabilized Lipases for Efficient Pancreatin Replacement Therapies

Enzyme replacement therapies (ERT) have been of great help over the past 30 years in treatment various lysosomal storage disorders, including chronic pancreatitis and its common complication, exocrine pancreatic insufficiency. Research shows that difficulties designing such drugs can be overcome by using appropriate additives enzyme immobilization techniques. Cyclodextrins (CDs) considered as a promising additive for therapies, they are known to enhance activity enzymes complex process due their specific binding. In this study, we investigated formulation lipases (from Aspergillus oryzae Burkholderia cepacia) paired with different cyclodextrins poly(vinyl alcohol) (PVA) nanofibers electrospinning technique. We examined effect presence nanoformulation on lipase activity. The rheological morphological characterizations precursors were also performed viscometer well electron Raman microscope. found selecting CD:lipase ratio, could multiplied, support homogeneous dispersion inside solid formula. addition, entrapment PVA led significant increase compared preformulated precursor. way, nanofibrous combining CDs provide an efficient sustainable possibility novel medicines ERT.
